ABSTRACT

Objective@#To compare the diagnosis and treatment experience of brain abscesses and improve prognosis.@*Methods@#The data of 302 patients of brain abscess at Department of Neurosurgery in Tianjin Medical University General Hospital from 1980 to 2014 was analyzed retrospectively. There were 215 male and 87 female patients aged from 11 to 82 years with mean age of (30±8) years. The patients was divided into 1980-2001 group and 2002-2014 group according to different diagnosis and the treatment methods. The therapy methods include operation and conservative treatment. There were 196 cases received operation, including 95 cases of excision, 89 cases of ventriculopuncture, 12 cases of excision after ventriculopuncture, 106 cases received drug conservative therapy. Two groups of information including clinical manifestation, abscess location, therapeutic effect and prognosis were compared by χ2 test.@*Results@#Compared to 1980-2001 group, adjacent infection incidence declined(χ2=8.000, P=0.005). The ratio of single abscess declined and multiple abscess increased(χ2=11.060, P=0.001), the infection proportion of frontal lobe and temporal lobe decreased(χ2=9.080, P=0.003; χ2=15.440, P=0.000). The ratio of headache and vomit and papilledema declined significantly(χ2=23.290, P=0.000; χ2=21.020, P=0.000; χ2=2.290, P=0.001). Total mortality of 302 patients were 23 cases and 5 cases of 1980-2001 group and 2002-2014 group (10.4% vs. 6.3%, χ2=1.180, P=0.277). However, there were statistical difference in postoperative mortality between both groups (14.4% vs. 4.0%, χ2 =3.880, P=0.049).@*Conclusion@#With the application of antibiotics and the development of neurosurgical techniques, the prognosis of brain abscess has been improved.

ABSTRACT

Objective To compare and analyze the occurrence of delayed cerebral ischemia(DCI)after coiling and clipping of intracranial aneurysms, and explore the risk factors of DCI. Methods A total of 236 patients with aneurysms diagnosed by CT angiography (CTA) or digital subtraction angiography (DSA) in Department of Neurosurgery, Tianjin Medical University General Hospital were enrolled in this study from March 2011 to May 2014. Patients were divided into clipping group(n=135) and coiling group(n=101). The clinical characteristics were compared between two groups, including gender, age, medical history, GCS score, Hunt-Hess grade, Fisher grade, WFNS grade, aneurysm location, prognosis and incidence of DCI. Risk factors for DCI were investigated by Logistic regression analysis. Results DCI was occurred in 36 patients (26.7%) underwent clipping operation while in 11 patients (10.9%) underwent coiling operation. The incidence was significantly higher in clipping group compared with that of coiling group (P 0.01). The overall mortality was 11.0%, the former had a lower mortality rate (5.9% vs. 17.8%, P <0.01). According to Logistic regression analysis, Fisher Grade 3-4, postoperative pulmonary infection and surgical procedure were independent risk factors for DCI (P<0.01). Conclusion DCI is one of the most significant factors for high fatality and morbidity of postoperative aneurysm patients. There is a low occurrence of DCI after coiling compared with that of clipping. If we pay more attention to risk factors associated with the DCI, it will improve the prognosis of postoperative aneurysm patients greatly.

ABSTRACT

Aim To evaluate the hypolipidemic effect of total phenylpropanoid glycoside extracted from Ligustrum robustum(Roxb.)Blume(CNTG)and its mecha-nisms.Methods The 60 hamsters were randomly divided into six groups,namely the control group,the model group,the positive control group(fenofibrate,150 mg·kg-1),the high(1 200 mg·kg-1),medium(600 mg·kg-1)and low(300 mg·kg-1)doses of CNTG groups.Only the control group was given control diet and other groups received high-fat diet.The changes of serum lipid were measured and analyzed in 1st week to ensure the successful establishment of the model.The drugs were administered daily for four weeks and the concentrations of lipids were determined in the 2nd week,3rd week and 4th week respectively.Quantitative real-time PCR and Western blot were used to assay the mRNA and protein expression of related signaling enzymes and proteins.Results Compared with the model group,the concentrations of serum TG,TC,LDL-C(P<0.05,P<0.01)and hepatic TG,TC(P<0.01)were effectively reduced in hamsters in CNTG-treated groups.Mechanism research found that CNTG increased the levels of phospho-AMPKα,LKB1 and phospho-LKB1 in liver(P<0.05).Conclusion CNTG prevents hyperlipidemia via activation of hepatic LKB1-AMPK pathway.

ABSTRACT

Abrin is isolated from abrin seeds (Aburs precatoriusL.), which is a kind of cytotoxic protein. This article reviewed the anticancer effect and mechanisms of Abrin from five aspects in several years, which were the molecular structure and toxicity of Abrin, the characteristics of Abrin in anticancer effect, the role of Abrin in inhibiting cancer cell proliferation, the mechanisms of Abrin induced apoptosis of cancer cells, the anticancer mechanisms of Abrin in enhancing immune function. And the application prospect of Abrin immunotoxin targeted therapy and nano preparation will be expounded in this article. It will be helpful to further research about application on Abrin. It will provide the scientific basis for discovering new drug targets of cancer.

ABSTRACT

Aim To establish the hyperlipidemic model and ex-plore the mechanism of hypercholesterolemia in hamster. Meth-ods Hamsters were randomly divided into the control and model groups. The hamsters in the control group were fed with the standard chow and the model group were fed with the high fat di-et. Serum lipids and CYP7A1 activity were detected by enzymat-ic method. The molecular mechanism of cholesterol metabolism was investigated by real-time PCR. Results In comparison with the control group, the concentrations of serum TC, LDL-C, TG and hepatic TC, TG were significantly increased in the model group. The mechanism research showed that in hamsters fed with the high fat diet, the CYP7A1 activity and the mRNA expres-sions of hepatic LDL-R, SREBP-2, CYP7A1, LXRαwere down-regulated, the expression of hepatic FXR was up-regulated. Conclusion The hyperlipidemic model could be developed in hamsters fed with the high fat diet for 4 weeks. LDL-R, SREBP-2, CYP7A1, FXR and LXRαare the biomarkers of hypercholes-terolemia, and also the targets of hypolipidemic drugs.

ABSTRACT

Aim To establish hyperlipidemic model and study the molecular mechanism of triglyceride (TG)disorder in hamsters.Methods The male ham-sters were randomly divided to control group fed with standard diet and model group fed with high-fat diet, both of the groups had been fed with diet for 4 weeks. The levels of serum TG,TC,LDL-C,FFA were detec-ted at the end of 2nd and 4th week.The hepatic TG, TC,LPL activity were detected by enzymatic method at the end of 4th week.The molecular mechanism was tested by real-time PCR.Meanwhile the effect of posi-tive drug fenofibrate on the model of hyperlipidemia in hamsters was investigated.Results Compared with the control,the serum levels of TG,TC,LDL-C,FFA in the model group increased 2.57,1.93,2.49,1.25 times at the end of2nd week,and 3.93,1.90,2.27, 2.29 times at the end of 4th week,respectively.The positive drug significantly decreased the concentrations of serum TG and FFA. The mechanism research showed that the hepatic AMPK,PPARα,CPT-1 mRNA decreased in hamsters fed with high-fat diet,and the SREBP-1 c,ACC,SCD-1 ,AGPAT2,DGAT2 mRNA ex-pressions increased.The hepatic ApoB mRNA expres-sion was up-regulated while the MTTP and LPL mRNA expressions were down-regulated slightly.LPL activity significantly decreased in model hamsters compared with the control.The alternations of these enzymes and receptors were the critical factors for TG disorder. Conclusion The hamsters fed with high-fat diet for 4 weeks can form a good hyperlipemic model with HTG feature.AMPK,SREBP-1 c,ACC,SCD-1 ,DGAT2,AG-PAT2,PPARα,CPT-1 and LPL are not only the main mechanisms of TG disorder,but also the biomarkers of hypotriglyceridemic drugs.

ABSTRACT

Object To study the effects of active fraction L.F04 from the ground part of Lycopus lucidus Turcz. var. hirtus Reg. on platelet aggregation and thrombosis formation and investigate its mechanisms of promoting blood circulation and removing blood stasis. Methods The effects of L.F04 on platelet aggregation induced by ADP in vivo, thrombosis of artery vein side road and thrombus formed in rotary loop in vitro were examined, the rat model of blood stasis made by injecting high molecular weight dextran (HMWD) was used. Results L.F04 0.408 and 0.204 g/kg evidently inhibited the ADP induced increase of platelet maximum aggregation rate in vivo in HMWD model, with a concentration dependent manner. Compared with the control group, the thrombus weight in rat model of blood stasis was increased significantly and the length of thrombus was shown an increasing trendency. L.F04 0.408 and 0.204 g/kg both shown the anti thrombosis effect. L.F04 0.408 g/kg showed better effects of lessening the thrombus dry weight and wet weight. Both L.F04 0.408 and 0.204 g/kg could inhibit the thrombosis of artery vein side road, the inhibition rates are separately 27.41% and 27.14%. Conclusion L. lucidus var. hirtus F04 could significantly inhibit platelet aggregation and thrombosis formation.

ABSTRACT

Lycopus lucidus is a herbal drug that can invigorate blood circulation,resolve blood stasis and improve microcirculation. In our further study of its pharmacological function and to identify its effective components,its effect on the hemorrheology of rat was used as a guide to evaluate the relative strength of four different extraction fractions of the drug. Results showed that fraction L. F04 and L. F02 were effective in reducing the blood viscosity,with L. Fo4 showing the best effect. This function of L. F04 was believed to be related to its ability to inhibit platelet aggregation and to improve the compostion of plasma

ABSTRACT

A model of s bacute senility state was established in mice by successive retrobulbar injectiou of 80 mg/kg D-galactose for 50 days. Concomitant oral administration of 4 g/kg and 8 g/kg gastrodia can effectively recovcr the decreased ability of the passive avoid ance reaction in the model animals.The activity of RBC SOD and the contents of hydroxyproline in the animals skin were markedly increased as well as the lipofuscin of myocardium. However, gastrodia was less effective to lower lipofuscin in liver and brain.
